Clinical Pharmacology & Mechanism of Action
CJC-1295 is a synthetic analogue of growth hormone-releasing hormone (GHRH). It is a tetrasubstituted 29-amino acid peptide that mimics the natural action of endogenous GHRH.
Its primary mechanism of action is to bind to GHRH receptors on the anterior pituitary gland, signaling it to release large pulses of natural Growth Hormone (GH) into the bloodstream .
Importantly, CJC-1295 does not replace your body's natural GH. Instead, it acts as a "secretagogue," encouraging your pituitary to produce more of its own.
This maintains the natural pulsatile rhythm of GH release, preventing the pituitary shutdown associated with synthetic HGH injections.
The DAC vs. No DAC Distinction
There is immense confusion regarding the naming convention of this peptide:
- CJC-1295 No DAC (Modified GRF 1-29): This version lacks the Drug Affinity Complex. It has a very short half-life of roughly 30 minutes, producing a massive but brief spike in GH.
- CJC-1295 with DAC: The addition of the Drug Affinity Complex allows the peptide to bind to serum albumin in the blood. This extends the half-life from 30 minutes to up to 8 days, causing a slow, constant bleed of GH rather than a sharp pulse.
Pharmacokinetics & Dosing Synergies
CJC-1295 is almost never researched in isolation. It only acts on one half of the pituitary's GH release mechanism.
To maximize GH output, researchers must simultaneously suppress Somatostatin (the hormone that inhibits GH release).
Therefore, CJC-1295 is universally stacked with a Growth Hormone Releasing Peptide (GHRP), most commonly Ipamorelin. When GHRH (CJC-1295) and GHRP (Ipamorelin) are administered together, the GH release is not just additive, it is exponentially synergistic.
Standard Research Dosing Protocols
Because "No DAC" is the standard for anti-aging and recovery research (to maintain natural GH pulses without GH bleed side effects), the following applies to CJC-1295 No DAC:
- Dosage: 100mcg per injection.
- Frequency: 1 to 3 times daily (usually morning, post-workout, and before bed).
- Administration Condition: Must be administered on an empty stomach (at least 2 hours after a meal containing carbohydrates or fats), as insulin blunts GH release.
Reconstitution Math & Syringe Guide
CJC-1295 is highly potent and dosed in very small microgram (mcg) quantities.
Step-by-Step Reconstitution (2mg Vial)
- Swab the rubber stoppers of your 2mg CJC-1295 vial and your Bacteriostatic Water.
- Using a sterile mixing syringe, draw exactly 2mL of Bacteriostatic Water.
- Inject the water into the 2mg vial slowly, aiming at the glass wall.
- Swirl gently to dissolve.
The Math for a Standard Dose (100mcg):
- 2mg total in the vial = 2,000mcg.
- You added 2mL of water.
- Therefore, the concentration is 1,000mcg per 1mL.
- You need a 100mcg dose. Divide the dose by the concentration: 100 ÷ 1,000 = 0.1mL.
- 0.1mL = 10 units on a U-100 syringe.
Common Mistakes & Troubleshooting
- Eating Food Before Injection: Administering CJC-1295 while blood glucose or insulin levels are elevated renders the peptide practically useless. A strict fasting window is required.
- Buying "With DAC" by Mistake: Constant GH bleed from DAC can lead to side effects like carpal tunnel syndrome, water retention, and severe lethargy. Always double-check which version you are purchasing.
- Shaking the Vial: GHRH peptides are fragile. Vigorous shaking will destroy the peptide chains.
