Clinical Pharmacology & Mechanism of Action
GHRP-6 (Growth Hormone Releasing Hexapeptide) is a synthetic, first-generation Growth Hormone Releasing Peptide (GHRP). It acts as a potent agonist of the ghrelin/growth hormone secretagogue receptor (GHSR).
Like all GHRPs, its primary function is to signal the anterior pituitary gland to release a massive pulse of stored Growth Hormone while simultaneously suppressing the release of somatostatin (the body's natural "stop" signal for GH) .
However, GHRP-6 is unique because of its overwhelming affinity for ghrelin pathways. Ghrelin is the hormone that signals the brain that the stomach is empty.
As a result, GHRP-6 induces intense, often uncontrollable hunger and gastric motility shortly after injection. Due to this severe appetite stimulation, it is rarely used for anti-aging or fat loss; instead, it is highly favored by researchers studying cachexia, muscle wasting, and extreme bulking in athletic contexts.
Pharmacokinetics & Systemic Responses
- Half-life: Roughly 15 to 30 minutes. The GH pulse clears the system in roughly 2-3 hours.
- Side Effects (Cortisol & Prolactin): Of all the GHRPs, GHRP-6 causes the most significant elevation in cortisol (stress hormone) and prolactin levels. It is the "dirtiest" GHRP, but many researchers tolerate these side effects in exchange for the profound appetite stimulation.
Standard Research Dosing Protocols
GHRP-6 is always stacked with a GHRH (like CJC-1295 No DAC) to maximize the GH pulse, though many use it solely for the hunger response.
- Dosage: 100mcg to 150mcg per injection.
- Frequency: 1 to 3 times daily (or 20 minutes before a large meal).
- Fasting Requirement: Must be injected on an empty stomach. Eating carbohydrates prior to injection completely blunts the GH release, even though the hunger side effect will still occur.
Reconstitution Math & Syringe Guide
GHRP-6 is highly potent and typically sold in 5mg vials. Precise reconstitution math is required to accurately dose in micrograms (mcg).
Step-by-Step Reconstitution (5mg Vial)
- Swab the rubber stoppers of your 5mg GHRP-6 vial and Bacteriostatic Water.
- Using a sterile mixing syringe, draw exactly 2mL of Bacteriostatic Water.
- Inject the water slowly down the inside wall of the GHRP-6 vial.
- Swirl gently.
The Math for a Standard Dose (100mcg):
- 5mg total in the vial = 5,000mcg.
- You added 2mL of water.
- Therefore, the concentration is 2,500mcg per 1mL.
- You need a 100mcg dose. Divide the dose by the concentration: 100 ÷ 2,500 = 0.04mL.
- 0.04mL = 4 units on a U-100 syringe.
(To make the math easier and draw 8 units instead of 4, you can add 4mL of BAC water to the vial, making the concentration 1,250mcg/mL).
Common Mistakes & Troubleshooting
- Eating Immediately After Injection: The intense hunger hits roughly 15-20 minutes after injection. If researchers eat carbohydrates immediately to satisfy the hunger, the resulting insulin spike destroys the GH pulse. You must endure the hunger for at least 30 minutes to allow the GH to peak before eating.
- Over-dosing: Injecting massive doses (e.g., 500mcg) does not produce more GH due to pituitary saturation. It only amplifies the cortisol and prolactin spikes, leading to water retention, lethargy, and gynecomastia risks.
- Drawing Errors: 4 units is extremely small. Using low-quality syringes with large dead space can result in highly inaccurate doses.
