Clinical Pharmacology & Mechanism of Action
Ipamorelin is widely considered the safest, mildest, and most targeted Growth Hormone Releasing Peptide (GHRP) available in clinical research. It is a synthetic pentapeptide that acts as a selective agonist of the ghrelin/growth hormone secretagogue receptor (GHSR) .
Unlike older GHRPs (like GHRP-2 or GHRP-6), Ipamorelin is unique because of what it doesn't do. Older GHRPs often cause severe hunger (by mimicking ghrelin too closely) and stimulate the release of cortisol (the stress hormone) and prolactin (which can cause gynecomastia).
Ipamorelin is highly selective: it stimulates a massive pulse of Growth Hormone from the pituitary gland without significantly elevating cortisol, prolactin, or intense hunger pathways.
Pharmacokinetics & Systemic Half-Life
Ipamorelin is a short-acting peptide designed to mimic the body's natural pulsatile rhythm of GH release.
- Half-life: Approximately 2 hours.
- Duration of Action: The GH pulse peaks roughly 40-60 minutes post-injection and returns to baseline within 3-4 hours.
Because of this short half-life, Ipamorelin is dosed multiple times a day to simulate the natural, youthful pattern of GH spikes, which is critical for lipolysis (fat burning) and cellular repair without shutting down natural pituitary function.
Standard Research Dosing Protocols
Ipamorelin is almost exclusively stacked with a GHRH (like CJC-1295 No DAC) to achieve a synergistic, exponential GH pulse.
- Dosage: 100mcg to 300mcg per injection.
- Saturation Point: Ipamorelin has a strict biological saturation point. Injecting more than 300mcg at one time yields diminishing returns. A 500mcg dose will barely produce more GH than a 300mcg dose.
- Frequency: 1 to 3 times daily. (Before bed is the most critical dose, as it aligns with the body's largest natural GH pulse).
Reconstitution Math & Syringe Guide
Ipamorelin is usually sold in 2mg or 5mg vials. Because the dose is in micrograms, calculation errors are common.
Step-by-Step Reconstitution (2mg Vial)
- Swab the rubber stoppers of your 2mg Ipamorelin vial and your Bacteriostatic Water.
- Using a sterile mixing syringe, draw exactly 2mL of Bacteriostatic Water.
- Inject the water slowly down the inside wall of the Ipamorelin vial.
- Swirl gently to dissolve the lyophilized powder.
The Math for a Standard Dose (200mcg):
- 2mg total in the vial = 2,000mcg.
- You added 2mL of water.
- Therefore, the concentration is 1,000mcg per 1mL.
- You need a 200mcg dose. Divide the dose by the concentration: 200 ÷ 1,000 = 0.2mL.
- 0.2mL = 20 units on a U-100 syringe.
Common Mistakes & Troubleshooting
- Injecting with Insulin Present: Like all GH secretagogues, insulin blunts the mechanism of action. If you eat a meal with carbohydrates or fats and then inject Ipamorelin, the GH pulse will be severely blunted. You must inject on a fasted stomach (2-3 hours after eating) and wait at least 30 minutes before eating again.
- Over-dosing: Because 10 units looks so small in a syringe, researchers often draw 50 units (500mcg) thinking more is better. Due to the saturation point, this simply wastes expensive peptide.
