Clinical Pharmacology & Mechanism of Action
Sermorelin is a synthetic 29-amino acid peptide that represents the shortest fully functional fragment of endogenous human Growth Hormone-Releasing Hormone (GHRH). Originally developed and FDA-approved in the 1990s as a diagnostic tool for pituitary function, it has become one of the most widely researched peptides for anti-aging and restoring youthful growth hormone levels.
It acts directly on the pituitary gland, binding to the GHRH receptor to stimulate the production and secretion of Growth Hormone . Because it relies on the body's natural pituitary feedback loop (somatostatin), Sermorelin is incredibly safe.
It cannot cause acromegaly or organ enlargement (unlike synthetic HGH abuse) because the body will naturally halt GH production if levels become too high.
Pharmacokinetics & Anti-Aging Applications
Sermorelin has a very short half-life (around 10-20 minutes). It enters the bloodstream, delivers the chemical "message" to the pituitary to release GH, and is rapidly broken down by enzymes.
This rapid clearance is highly desirable in anti-aging protocols because it exactly mimics how the body naturally pulses GH. As we age, our natural GH pulses diminish in amplitude. Administering Sermorelin restores the amplitude of these pulses to youthful levels, which aids in:
- Deepened slow-wave sleep (REM sleep)
- Improved skin elasticity and collagen synthesis
- Accelerated lipolysis (fat burning)
- Enhanced recovery from exercise
Standard Research Dosing Protocols
Because Sermorelin is slightly less potent milligram-for-milligram than newer GHRH analogues (like CJC-1295), standard research doses are slightly higher.
- Dosage: 200mcg to 300mcg per injection.
- Frequency: Once daily, typically administered immediately before bed.
- Timing: Must be administered on an empty stomach to avoid insulin blunting.
Reconstitution Math & Syringe Guide
Sermorelin is notoriously difficult to calculate because it often comes in non-standard vial sizes (like 9mg) from compounding pharmacies, or 5mg vials from research chemical suppliers.
Step-by-Step Reconstitution (5mg Vial)
- Swab the rubber stoppers of your 5mg Sermorelin vial and Bacteriostatic Water.
- Using a sterile mixing syringe, draw exactly 2.5mL of Bacteriostatic Water. (We use 2.5mL here to make the math very clean).
- Inject the water slowly down the inside wall of the Sermorelin vial.
- Swirl gently to dissolve.
The Math for a Standard Dose (300mcg):
- 5mg total in the vial = 5,000mcg.
- You added 2.5mL of water.
- Therefore, the concentration is 2,000mcg per 1mL.
- You need a 300mcg dose. Divide the dose by the concentration: 300 ÷ 2,000 = 0.15mL.
- 0.15mL = 15 units on a U-100 syringe.
Common Mistakes & Troubleshooting
- Using the Wrong Water Volume: If you add 2mL of water to a 5mg vial, the math becomes complex (2.5mg/mL), and drawing exactly 300mcg requires drawing exactly 12 units. Adding 2.5mL of water simplifies the math significantly.
- Degradation from Temperature: Sermorelin is considered slightly less stable than CJC-1295 once reconstituted. It must be kept strictly refrigerated. Leaving the mixed vial on a counter overnight can result in a significant loss of potency.
- Expecting Immediate Weight Loss: Unlike GLP-1s, GH secretagogues take time to work. Improved sleep is often noticed in the first week, but physical changes (fat loss, skin improvement) generally require 3 to 6 months of consistent use.
