Mechanism of Action & Biochemical Context

Sermorelin is a synthetic 29-amino acid peptide representing the biologically active N-terminal fragment of human GHRH. It was FDA-approved as a diagnostic agent, and carries decades of clinical safety data for GH stimulation research.

Storage, Reconstitution & Stability Guidelines

Prior to reconstitution, lyophilized Sermorelin powder should be stored away from light between 2°C and 8°C for short-term access, or -20°C for long-term preservation. Once reconstituted with Bacteriostatic Water (0.9% benzyl alcohol preservative), maintain strict refrigeration between 2°C and 8°C. Do not freeze liquid solution.

Frequently Asked Questions

2.5mL of Bacteriostatic Water is the standard for a 5mg Sermorelin vial. This creates a clean, convenient concentration of 2,000mcg/mL (2mg/mL), making a 300mcg dose exactly 15 units (0.15mL) on a U-100 syringe - a well-proportioned, easy-to-draw volume.

200mcg to 300mcg, administered once daily before bedtime. Sermorelin's extremely short half-life (10-20 minutes) means it delivers a rapid pulse and then clears the system. The before-bed timing is critical as it aligns with the body's peak circadian GH pulse, producing a compounding effect on natural overnight GH secretion.

Sermorelin is the original, FDA-approved GHRH peptide with decades of clinical safety data. CJC-1295 is a modified, more potent version - its tetrasubstitution makes it more resistant to enzymatic degradation, resulting in a higher potency milligram-for-milligram and a slightly longer half-life (30 minutes vs 10-20 minutes). For clinical safety preference, Sermorelin is chosen; for maximum GH stimulation, CJC-1295 No DAC is typically preferred.

Stacking with a GHRP (like Ipamorelin or GHRP-2) dramatically amplifies the GH pulse, but Sermorelin does work in isolation. On its own, Sermorelin only addresses one part of the pituitary mechanism (the GHRH signal). Adding a GHRP suppresses somatostatin and directly stimulates the GH secretagogue receptor, resulting in a synergistic and significantly more robust GH release.

Because GH secretagogue effects build slowly (improved sleep quality is noticeable in week 1-2, but body composition and skin changes emerge over months), most research protocols run for a minimum of 3 months. Many researchers continue for 6 months, followed by 1-2 month breaks. Longer cycles are feasible because Sermorelin works within the natural feedback loop and does not shut down the pituitary.

Sermorelin is a relatively short peptide (29 amino acids) with a straightforward linear structure. However, it is still susceptible to enzymatic degradation in the bloodstream, which is why its half-life is short and why refrigerated storage of reconstituted vials is mandatory. It is generally considered stable for 28-30 days in the fridge after reconstitution with BAC water.

In research subjects, common documented outcomes over 3-6 months include: significantly improved slow-wave (deep) sleep quality, reduction in abdominal fat mass, improved skin elasticity and reduced wrinkle depth, increased lean muscle mass, improved recovery from exercise, and elevated energy levels and mood. These effects are more gradual and subtle than synthetic HGH but are safer and more sustainable.
